چکیده

FTIPPURIC ACID 1S an important metabolic product normally present in urine in amounts determined by tile ingestion of benzyl-containing foods and medicinals (ii). Because this compound is comparatively inert chemically, identification requires isolation and the determination of physical constants after extensive purification. There has long been need for a reliable and sensitive color reaction for its detection and for estimation of low-level concentrations. In attempts to enhance chromatographic separations of components in acidic extracts of urine through preparation of benzoyl and related derivatives, development of a deep-red color with benzenesulfonyl chloride (BSC) in the presence of pyridine (Py) was traced to hippuric acid (HA) in the mixture. Studies of the reaction led to a sensitive color test applicable for quantitative measurements. Other alkyl and aryl acid halides failed to develop significant colors and amino acids and related acidic compounds did not react in the test. Salicyluric acid is equally sensitive and probably other amido acids are a source of interference. Confirmation of HA is readily made following positive findings for an amido structure in the test.

Comparison of several methods for the measurement of urinary hippuric acid as an index of toluene exposure.

The levels of hippuric acid in the urine of people exposed to toluene vapour were measured by paper chromatography, direct colorimetry, high performance liquid chromatography and gas chromatography. The control was a similar group not exposed to toluene vapour. The values were analyzed statistically, conversion equations calculated, and the propriety of these equation discussed. Since the three...

Direct colorimetric determination of hippuric acid in urine.

In 1963 Umberger and Fiorese (1) introduced a colorimetric method for HA,1 based on the color reaction of HA, dissolved in anhydrous pyridine, with BSC. Therefore, for applying their method to determine HA in urine, HA must first be extracted from the urine. We found that HA dissolved in water-pyridine (1:1, by vol) produces a red-orange color when BSC is added at room temperature. The absorpti...

An improved direct colorimetric method for the quantitative analysis of urinary hippuric acid as an index of toluene exposure.

An improved direct colorimetric method for determining the concentration of urinary hippuric acid as an index of toluene exposure was described. One tenth ml of urine was diluted with 0.4 ml 0.01 M phosphate buffer H 6.9 and mixed with 0.5 ml pyridine. The mixture was layered on 0.2 ml benzenesulfonyl chloride. The reaction was started by mixing for one min with a mechanical shaker. The colored...

Quantitative determination of hippuric and m-methylhippuric acids in urine by high-speed liquid chromatography.

High-speed liquid chromatography employing an ultraviolet photometric detector has been applied to the simultaneous determination of hippuric and m-methylhippuric acids in urine. Reversed-phase partition chromatography is carried out on a muBondapak C18 column with methanol-water as the eluent system. This method obviates the necessity for isolation or reaction of these acids before assay. The ...

Colorimetric determination of hippuric acid in urine and liver homogenate.

To a dried extract Containing hippuric acid (HA), 1.0 mL of acetic anhydride and 2.0 mL of 0.5% p-dlmethyiamlnobenzaldehyde (DAB) solution In pyridine were added, and the solution was kept at 40 O C for 1 h after thorough mlxing. The absorbance was then determlned at 458 nm agalnst a blank containing acetic anhydride, DAB, and pyridine. This method was in good agreement with Beer’s law within 1...

Sensitive colorimetric assay for angiotensin converting enzyme in serum.

A sensitive colorimetric procedure has been developed for the assay of angiotensin converting enzyme (EC 3.4.15.1) in serum. Serum (10 microL) is incubated for 30 min with hippuryl-glycyl-glycine as described earlier (Clin Chem 28: 1352-1355, 1982). After a Folin-Wu deproteinization, the liberated glycyl-glycine is derivatized with a borate-buffered (pH 9.3) trinitrobenzenesulfonate solution (6...
